Birth. June 1, 1630, Rome. Nephew of Cardinals Francesco Barberini, seniore (1623), and Antonio Barberini, iuniore, O.S.Io.Hieros. (1627). Grand-nephew of Pope Urban VIII (1623-1644) and of Cardinal Antonio Barberini, seniore, O.F.M.Cap. (1624). Uncle of Cardinal Francesco Barberini, iuniore (1690). Called to the ecclesiastical life, resigned his primogeniture in favor of his younger brother Maffeo.

Cardinalate. Created cardinal deacon in the consistory of June 23, 1653; with dispensation for having two uncles in the Sacred College of Cardinals; received the red hat and the deaconry of S. Cesareo in Palatio, August 18, 1653. At his promotion to the cardinalate, he received the faculty of retaining the charge of prefect of Rome (1). Participated in the conclave of 1655, which elected Pope Alexander VII. Opted for the deaconry of S. Angelo in Pescheria, August 30, 1660. In the absence of his uncle Cardinal Antonio Barberini, was elected administrator of the patriarchal Liberian basilica and of the diocese of Palestrina. Participated in the conclave of 1667, which elected Pope Clement IX. Named archpriest of the patriarchal Vatican basilica when his uncle Francesco Barberini resigned the post, and also pro-camerlengo of the Holy Roman Church during the absence of his uncle Antonio Barberini. Participated in the conclave of 1669-1670, which elected Pope Clement X. Opted for the deaconry of S. Cesareo in Palatio again, November 14, 1667. Opted for the deaconry of S. Maria in Cosmedin, December 2, 1675. Participated in the conclave of 1676, which elected Pope Innocent XI. Opted for the order of priests and the title of S. Maria della Pace, September 27, 1683. Opted for the title of S. Lorenzo in Lucina, April 30, 1685. Cardinal protoprete. Participated in the conclave of 1689, which elected Pope Alexander VIII. Participated in the conclave of 1691, which elected Pope Innocent XII. Prefect of the S.C. of Propaganda Fide, July 17, 1698 until his death. Participated in the conclave of 1700, which elected Pope Clement XI. Legate a latere to Naples to compliment King Felipe V of Spain in the name of the pope, 1702.
Death. October 2, 1704, Rome. Exposed in the church of S. Andrea della Valle, Rome, and buried near the altar of the Barberini chapel in that same church.
